Abstract: 目的 探讨使用特殊设计的单链寡核苷酸链(ODN)纠正抗肌萎缩蛋白基因的点突变.方法 给予DMDmdx小鼠肌内注射低剂量和高剂量的ODN,以及另外附加透明质酸酶预处理和活体电穿孔术,采用扩增阻滞突变体系(ARMS)法检测点突变纠正情况.结果 不管是低剂量和高剂量ODN肌内注射,还是在此基础上另外再附加电穿孔术,都看不到纠正了的基因条带.结论 使用的实验方法不能纠正抗肌萎缩蛋白基因的点突变或纠正率过低ARMS法检测不出.
