Abstract: 试验采用SD大鼠作为试验动物,通过以高脂饲料诱导其高脂血症地鼠作为动物模型,研究单方中药地骨皮中蒽醌成分对高脂血症SD大鼠胆汁酸代谢的调节作用,阐明地骨皮对调节高脂血症大鼠胆汁酸的有效成分的影响及其机理.雄性SD大鼠适应性喂养1周后,随机分为正常对照组(10只,喂饲普通饲料)和模型组(50只,喂饲高脂饲料).将造模成功大鼠40只随机分为辛伐他汀组、地骨皮蒽醌低、高剂量组,每组10只.大鼠单笼单只养殖,在0、1、2月,各收集4d粪便标本,以乙醇-KOH抽提,以酶法测定胆汁酸及胆固醇浓度.试验前大鼠粪便胆汁酸各组之间无显著差异(P>0.05);药物干预后,蒽醌高、低剂量组与模型组差异极显著(P<0.01),辛伐他汀组与模型组比无显著性差异(P>0.05).试验结果表明,和模型组比较,地骨皮总蒽醌显著升高高脂大鼠粪便中的胆汁酸水平,并且胆汁酸升高水平与地骨皮总蒽醌的给药剂量呈正相关.表明地骨皮总蒽醌的降脂机理可能与增加胆汁酸的外排有关.
