Determination of eight carbohydrates in juices and dairy beverages by hydrophilic interaction chromatography

Abstract: [Objective] To establish a hydrophilic interaction chromatography (HILIC) method for the determination of eight carohydrates in juices and dairy beverages.[Methods] The juices and dairy beverages samples were diluted by acetonitrile:water (50 ∶ 50) solution and separated on a HILIC Waters(R) Xbridge BEH Amide (4.6 mm×250 mm,5 μm) column,acetonitrile:water (75 ∶ 25) solution containing 0.2% (v/v) triethylamine was used as mobile phasethe,flow rate was 1.2 ml/min,and the column temperature was 50 C.8 carbohydrates (xylose,fructose,glucose,sucrose,lactose,maltotriose,isomaltotriose and stachyose) were quantitatively analyzed by differential refractive index detector.[Results] The correlation coefficients of standard curves for xylose,fructose,glucose,sucrose,lactose,maltotriose,isomaltotriose and stachyose in linearity were good in the ranges of 0.40-40.0,0.30-30.0,0.40-40.0,0.20-20.0,0.40-40.0,0.06-3.0,0.04-2.0 and 0.40-20.0 mg/ml respectively,as well as the recoveries of all the eight carbohydrates were 95.9%-105.1% with the relative standard deviations (RSD) of 0.03%-4.86% at three spiked levels.After about 350 hours (approximately 6 000 times the column volume of mobile phase) of continuous elution,the performance of the HILIC column did not change obviously.[Conclusion] The method is simple,accurate and repeatable.The technical parameters meet the requirements of physical and chemical analysis of food and health food.Compared with traditional NH2 column,HILIC column has better chemical stability which avoids column loss during use process,so that HILIC column has longer service life and better durability,and can be used for the test and quality control of the eight carbohydrates in juices and dairy beverages.
