Determination of 5 alkaloids in hot pot condiment by high performance liquid chromatography-electrospray tandem mass spectrometry

Abstract: [Objective] To establish a method for determination of morphine,codeine,papaverine,narcotine and thebaine in hot pot condiment by high performance liquid chromatography-triple quadrupole tandem mass spectrometry (HPLC-MS/MS).[Methods] Samples were extracted with 1% trichloracetic acid,cleaned up by solid phase extraction cartridge,and eluted with 5% ammonia methanol solution.The eluent was blown to near dry with a nitrogen concentrator and dissolved in the 1 ml initial mobile phase.The separation of analytes was carried out on a C18 column,and the quantification analysis was achieved by HPLC-MS/MS using external standard method.[Results] The instrument parameters were optimized to effectively overcome various disturbances,and the method indicators,such as the precision,the linear range as well as the regression coefficients,were consistent with the standards.The detection limit of the method was 0.2 μg/kg,the relative coefficients were higher than 0.999,the recovery was found to be in the range of 62.8%-106.0%,and the relative standard deviation (RSD) was≤9.6%.[Conclusion] The method has the advantages of sample preparation,less extraction solvent consumption,short operation time and high efficiency,and can be widelv used in the determination of poppy shell alkaloids in hot pot soup,sauce and seasoning.
